Molecular Assays

Profile immune repertoires and track engineered cells in your samples

Are you interested in tracking expansion of cellular therapy products, or analyzing the diversity of T and B cell receptor repertoires in your study samples? HIMSR offers comprehensive molecular assay services designed to support your immune monitoring needs. Our team provides expertise in digital PCR, TCR sequencing, and BCR sequencing, working in close collaboration with the UCCC Functional Genomics, Flow Cytometry, and Genomics Shared Resources to ensure high-quality results.

RNA/DNA isolation

We offer RNA and DNA isolation from a variety of sources, including whole blood, fixed cells, PBMCs, and FFPE tissue.

RNA quality control

Our services include the DV200 assay for RNA quality assessment or RNAscope assay for spatial transcriptomics.

Digital PCR

Digital PCR enables highly sensitive and precise detection of rare sequences, making it an ideal platform to monitor CAR T cell persistence and expansion in patient samples following adoptive transfer. HIMSR provides full-service workflows for assay development, sample preparation, and data analysis to ensure reliable and reproducible detection, utilizing the QIAcuity instrument, available at the Functional Genomics Shared Resource.

QIAcuityDx Four Instrument

Bulk repertoire analysis (TCR and BCR sequencing)

For profiling T cell receptor repertoires, we utilize for genomics DNA. HIMSR supports this service by performing sample preparation, optional cell sorting using instruments in the Flow Cytometry Shared Resource, genomic DNA extraction, and library preparation. Kits for TCR alpha, beta, delta, gamma, and BCR IgH and L are also available for RNA. Sequencing is carried out in collaboration with the Genomics Shared Resource, and online analysis tools are available through iRepertoire.

Single cell clonality analysis

For characterization of T cell and B cell receptor clonotypes, HIMSR can provide end-to-end sample preparation and cell sorting (in collaboration with the Flow Cytometry Shared Resource prior to sequencing. Cells are transferred to the Genomics Shared Resource for single-cell RNA sequencing (scRNAseq), enabling high-resolution insights into T cell and B cell diversity and clonality.
